Receptor activation in hypothalamic and limbic circuits is the mechanism most often cited for the observed effects on sexual desire. Signalling through MC4R couples to Gs proteins and raises intracellular cyclic AMP, which in turn modulates dopaminergic tone in reward-related pathways. Because the peptide reaches the central nervous system after subcutaneous administration, peripheral vascular changes are regarded as secondary rather than primary. The precise neural circuits that translate receptor occupancy into behavioural change remain incompletely mapped, and published accounts describe the pathway in general terms rather than as a fully resolved sequence.
Development began with intranasal formulations investigated for erectile dysfunction, but blood pressure elevation limited that route and prompted a switch to subcutaneous delivery. Clinical testing then shifted toward hypoactive sexual desire disorder in premenopausal women, and a subcutaneous product received United States approval in 2019. Later trials examined other populations with mixed results, and questions about effect size, durability and patient selection remain open in the peer-reviewed literature. Research interest continues in parallel with the broader melanocortin field, where several synthetic analogues are studied together.
PT-141 is the research code for bremelanotide, a cyclic heptapeptide derived from alpha-melanocyte-stimulating hormone. The molecule belongs to the melanocortin receptor agonist family and shows highest affinity for the MC4 receptor subtype, with weaker activity at MC1, MC3 and MC5. Its structure retains the core His-Phe-Arg-Trp sequence that defines melanocortin recognition, while cyclization and terminal modifications improve metabolic stability relative to the parent hormone. Early work classified the compound as a centrally acting agent rather than a peripherally acting vasodilator, which shaped subsequent development priorities.
Published studies differ in design, population and endpoint definition, so results are not always directly comparable across reports. Some trials used patient-reported measures of desire and distress, while others tracked physiological or behavioural outcomes. Questions that remain open include the durability of effects beyond short follow-up periods, the frequency of transient blood pressure elevation observed after administration, and whether a subtype-selective analogue could separate central effects from pigmentation-related activity. These points are usually framed as unresolved rather than settled in review literature.
Routine characterisation of bremelanotide relies on reversed-phase high-performance liquid chromatography with ultraviolet detection near 214 nm, where the peptide backbone absorbs. Mass spectrometry, typically in tandem mode with electrospray ionisation, confirms identity and supports quantification in biological matrices. Additional checks include amino acid analysis, peptide mapping after enzymatic digestion, and confirmation of the lactam bridge, since incomplete cyclisation produces a mass-shifted by-product. Purity values above 95 percent are common in reference-grade material, though reports vary in how strictly related substances are resolved from the parent peak.

Compared with melanotan II, bremelanotide is a smaller cyclic peptide with a more constrained backbone, which affects receptor selectivity and metabolic stability. Published descriptions give a plasma half-life on the order of a few hours after subcutaneous administration, with elimination through hepatic and renal routes and limited plasma protein binding. Central access is inferred from effects observed in animal models, although direct measurement in humans is limited. Handling and storage requirements follow from the peptide backbone, which is susceptible to hydrolysis and oxidation.
The melanocortin system comprises five G protein-coupled receptors, designated MC1 through MC5, that signal mainly through cyclic AMP accumulation. MC1R and MC2R are associated with pigmentation and adrenal steroid production, while MC3R and MC4R are expressed in the central nervous system and influence energy balance and behavior. MC5R appears in exocrine tissues. Natural agonists include alpha-melanocyte-stimulating hormone and adrenocorticotropic hormone, and endogenous antagonists such as agouti-related protein modulate the same sites. This receptor family provides the framework within which bremelanotide activity is described.
Handling procedures emphasize minimizing moisture uptake, since the dried solid is hygroscopic and can draw water when warmed to room temperature before a vial is opened. Laboratories record lot number, purity, counter-ion content, and residual solvent data supplied by the producer. Impurity profiles are compared run to run, and any shift in retention time or peak shape prompts re-verification against a reference standard. Certificates of analysis are treated as claims to be checked rather than accepted at face value.
Identity and purity testing for this peptide typically relies on reversed-phase high-performance liquid chromatography with ultraviolet detection, reported as area-percent purity. Mass spectrometry, usually in tandem mode, confirms molecular mass and supports quantification in biological matrices. Sequence confirmation may use peptide mapping after enzymatic digestion, while nuclear magnetic resonance and circular dichroism supply supplementary structural detail. No single technique establishes identity alone, so laboratories compare retention time, mass, and fragment pattern against a verified reference standard.
The lactam ring that closes the peptide backbone improves resistance to exopeptidase attack, but the molecule stays susceptible to hydrolysis and oxidation once dissolved. Degradation accelerates with temperature, extreme pH, and light exposure, and repeated freeze-thaw cycles promote aggregation and material loss. Lyophilized powder held desiccated at or below minus twenty degrees Celsius is the common way to keep reference material. Reconstituted solutions are generally kept cold and used within a short window because their stability is far lower than that of the dry solid.
Clinical development of bremelanotide proceeded through several reformulation attempts. An early intranasal version was discontinued, and a subcutaneous auto-injector formulation later received approval for hypoactive sexual desire disorder in premenopausal women. Approval decisions have varied by country and over time, and the product has not been universally adopted. Blood pressure elevation is a documented effect, which is why some jurisdictions require monitoring after administration. The clinical evidence base continues to evolve as additional studies are published.
Bremelanotide is a cyclic heptapeptide that acts as an agonist at melanocortin receptors. It binds MC1R, MC3R, MC4R, and MC5R, with MC4R activation considered most relevant to sexual desire pathways in the central nervous system. The molecule is a synthetic analog of alpha-melanocyte-stimulating hormone, a naturally occurring peptide involved in pigmentation and energy regulation. Early research explored its use in tanning before attention shifted toward sexual dysfunction applications. Receptor binding affinity varies across these subtypes.
Activation of MC4R in the hypothalamus is thought to influence dopaminergic signaling, which in turn affects arousal and desire. This mechanism differs from that of phosphodiesterase type 5 inhibitors, which act primarily on vascular smooth muscle in the genital region. Because the pathway is central rather than peripheral, effects are not strictly dependent on local blood flow. The precise downstream cascade linking receptor binding to behavioral outcomes remains an area of ongoing investigation.
The compound emerged from a research programme examining melanocortin analogues for effects on skin pigmentation. During early human studies, participants reported spontaneous erections as an unexpected side effect, which redirected development toward sexual function rather than tanning. An intranasal formulation was investigated in clinical trials but did not reach market approval. A subcutaneous injectable version later completed the regulatory process, and the nasal route does not appear in approved labelling.
Approved use is narrow and jurisdiction-specific. In the United States, the injectable product is authorised for premenopausal women with acquired, generalised hypoactive sexual desire disorder, a diagnosis that requires documented distress. It is not approved for men, for postmenopausal women, or for use alongside hormonal contraceptives under the approved labelling. Outside regulated markets, the same peptide is frequently sold as a research chemical, where identity, purity, and sterility are not independently verified.
Bremelanotide is a synthetic cyclic heptapeptide developed as an analogue of alpha-melanocyte-stimulating hormone, a naturally occurring peptide involved in pigmentation and appetite signalling. Its structure contains seven amino acid residues joined by a lactam bridge that closes the ring between two side chains. The molecular formula is C50H68N14O10 and the nominal molecular mass is near 1025 daltons. Much of the early laboratory literature refers to the same molecule by the development code PT-141.

With ring-opening polymerization, automated synthesis has been used for rapid screening and optimization, including with catalyst + initiator systems and their polymerization conditions. For example, Hoogenboom et al. determined the optimal temperature for the polymerization of 2-ethyl-2-oxazoline in dimethylacetamide (DMAc), allowing for individual heating of the parallel reactors, which shortened the time needed for preparation and analysis. To aid with the catalyst research for polyolefins, Symyx Technologies Inc. used automated synthesis to create a library of palladium and nickel catalysts, which were screened for ethylene polymerization. This process found that the largest polyethylene polymers were created by the complexes with the highest steric hindrance for the ortho-positions of the aryl rings, while electronic factors did not influence yield or molecular weight. In addition, Tuchbreiter and Mülhaupt used automated synthesis to demonstrate the improvements of minireactors for the polymerization of olefins, with quality improving as compared to utilizing simple arrays.

Gas exchange must be restored as quickly as possible to avoid collateral damage, so activated lymphocytes secrete IFNγ to stimulate the production of matrix metalloproteinase MMP-9 by macrophages. AMs have been reported to produce MMP-9 partly via PGE2-dependent PKA signaling pathways, which are the pathways involved in the inhibition of phagocytosis. MMP-9 activates latent TGF-β, reinducing expression of αvβ6 integrins on alveolar epithelial cells, thereby returning the alveolar macrophage to a resting state. Activation of TGF-β is also advantageous because its production stimulates collagen synthesis in interstitial fibroblasts, which is necessary for restoring alveolar wall architecture. The first National Air Pollution Symposium in the United States was held in 1949 and hosted by Stanford Research Institute (now SRI International). At first, smaller governments were responsible for the passage and enforcement of such legislation. The main purpose of the Air Pollution Control Act of 1955 was to provide research assistance to find a way to control air pollution from its source. A total of $5 million was granted to the public health service for a five-year period to conduct this research. According to a private website, the amount was $3 million allotted per year for the five-year period of research.
The main etiologic factor of ACC is unknown, although families with Li–Fraumeni syndrome, caused by an inherited inactivation mutation in TP53, have increased risk. Several genes have been shown to be recurrently mutated, including TP53, CTNNB1, MEN1, PRKAR1A, RPL22, and DAXX. The telomerase gene TERT is often amplified while ZNRF3 and CDKN2A are often homozygously deleted. The genes h19, insulin-like growth factor II (IGF-II), and p57kip2 are important for fetal growth and development. They are located on chromosome 11p. Expression of the h19 gene is markedly reduced in both nonfunctioning and functioning adrenal cortical carcinomas, especially in tumors producing cortisol and aldosterone. Also, a loss occurs of activity of the p57kip2 gene product in virilizing adenomas and adrenal cortical carcinomas. In contrast, IGF-II gene expression has been shown to be high in adrenal cortical carcinomas. Finally, c-myc gene expression is relatively high in neoplasms, and it is often linked to poor prognosis. Bilateral adrenocortical tumors are less common than unilateral. The majority of bilateral tumours can be distinguished according to size and aspect of the nodules: primary pigmented nodular adrenocortical disease, which can be sporadic or part of Carney complex, and primary bilateral macro nodular adrenal hyperplasia.Metastasis is most commonly to the liver and lung.
First observations of short-lived pear-shaped atomic nuclei Research conducting using the Miniball experimental setup found evidence of pear-shaped heavy nuclei, in particular radon-220 and radium-224. These results were named in the Institute of Physics (IoP) "top 10 breakthroughs in physics" in 2013, and was featured as the cover of Nature 2013. In 2020, due to the HIE-ISOLDE upgrade, radium-222 was also found to have a "stable pear shape". Laser spectroscopy has been performed on a short-lived radioactive molecule, containing radium, which further studies into could reveal physics beyond the Standard Model due to time-reversal symmetry breaking. Measurement of 229mTh transition energy In 2023, ISOLDE made the first 1%-level measurement of the ultralow-energy thorium-229m nuclear isomer, detecting photons at an energy of 8.338±0.024 eV. This was a key step in the construction of a future nuclear clock. Below is a list of improvements needed for the ISOLDE facility, considering both medium and long-term goals. Some of these improvements have been proposed by the EPIC project.
The T5 exonuclease chews back DNA from the 5' end of each fragment, exposing 3' overhangs on each DNA fragment. The complementary overhangs on adjacent DNA fragments anneal via complementary base pairing. The Phusion DNA polymerase fills in any gaps where the fragments anneal. Taq DNA ligase repairs the nicks on both DNA strands. Because the T5 exonuclease is heat labile, it is inactivated at 50 °C after the initial chew back step. The product is thus stable, and the fragments assembled in the desired order. This one-pot protocol can assemble up to 5 different fragments accurately, while several commercial providers have kits to accurately assemble up to 15 different fragments in a two-step reaction. However, while the Gibson assembly protocol is fast and uses relatively few reagents, it requires bespoke DNA synthesis as each fragment has to be designed to contain overlapping sequences with the adjacent fragments and amplified via PCR. This reliance on PCR may also affect the fidelity of the reaction when long fragments, fragments with high GC content or repeat sequences are used.

Immune evasion proteins from Staphylococcus aureus have a significant conservation of protein structures and a range of activities that are all directed at the two key elements of host immunity, complement and neutrophils. These secreted virulence factors assist the bacterium in surviving immune response mechanisms. Examples of toxins produced by strains of S. aureus include enterotoxins that cause food-poisoning, exfoliative toxins that cause scalded skin syndrome, and toxic-shock syndrome toxin (TSST) that underlies toxic shock syndrome. These toxin examples are classified as superantigens. Multi-drug resistant S. aureus strains also produce alpha toxin, classified as a pore-forming toxin, which can cause abscesses.
In the laboratory it is a common precipitant and cryoprotectant in protein crystallography. Since hexylene glycol is compatible with polar and nonpolar molecules, it competes with the solvent in a crystallography experiment causing the protein to precipitate. Hexylene glycol is so effective in protein crystallography because its amphiphilic nature and small, flexible structure allows it to bind to many different locations on a protein secondary structure including alpha helices and beta sheets. When hexylene glycol binds to these different locations, water is removed and the protein crystals anneal, which prevents ice formation during cryocrystallography techniques. Incorporation of hexylene glycol into solution has been known to improve the resolution of X-ray diffraction making protein structures easily identifiable. Additionally hexylene glycol is not a strong denaturing agent and thus does not significantly alter the structure of a protein during the crystallography procedure. Hexylene glycol is also used as a lubricant for polishing specimens in metallography. Like related diols, it forms borate esters.
One of the main roles of the INO80 subfamily is the incorporation and removal of alternate histones in the nucleosome. In the presence of the H2A.Z histone, the INO80 subfamily of remodelers catalyze the repositioning and eventual removal of this alternate histone. The H2A.Z histone is found on the first nucleosome at the beginning of genes. The INO80 subfamily of remodelers will also be recruited to the H2A.X histone in the homologous recombination repair pathway. In addition to this function, the INO80 subfamily plays a role in transcriptional regulation and genomic recombination. In the DNA damage pathways, the INO80 subfamily aids in repair, recombination, and cell cycle regulation. The INO80 subfamily can activate the recruitment of replication checkpoint factors and can aid in the recovery from replicative stress on the DNA strand. The subfamily's ability to incorporate alternate histones is important for genome stability, disease pathogenesis, and stem cell identity. INO80 complexes commonly bind to nucleosome free regions at transcription start sites and termination sites. INO80 is the only remodeler that is able to use the energy from ATP hydrolysis to create nucleosome free regions and cooperate with other remodelers to equally space nucleosomes.
Adenomatous polyposis coli protein 2 is a protein that in humans is encoded by the APC2 gene. This is a dual-functional protein which is especially prevalant in the developing brain, and involved in cell signaling (specifically Wnt signaling) and stabilizing the cytoskeleton. Frizzled receptors are a type of cell receptor involved in a variety of functions including embryonic development and cell proliferation. These receptors trigger a cascade of reactions called the Wnt signaling pathway that eventually leads to interaction with the DNA via the protein beta-catenin, which affects which proteins get produced by the cell. The APC2 protein promotes the formation of a protein complex which is involved in breaking down the protein beta-catenin. The APC2 protein stabilizes microtubules, which are part of the cell cytoskeleton. It may also be involved in the regulation of actin fibers my means of GTPase activation (specifically the Rho family).
4-Aminobenzoic acid (also known as para-aminobenzoic acid or PABA because the two functional groups are attached to the benzene ring across from one another in the para position) is an organic compound with the formula H2NC6H4CO2H. PABA is a white crystalline solid, although commercial samples can appear gray. It is slightly soluble in water. It consists of a benzene ring substituted with amino and carboxyl groups. The compound occurs extensively in the natural world. In industry, PABA is prepared mainly by two routes: Reduction of 4-nitrobenzoic acid Hoffman degradation of the monoamide derived from terephthalic acid. Food sources of PABA include liver, brewer's yeast (and unfiltered beer), kidney, molasses, mushrooms, and whole grains. Other food sources of PABA include spinach and oat seeds.

The code designates bremelanotide, a cyclic heptapeptide melanocortin receptor agonist. It served as an internal research identifier before the compound entered formal clinical development. The code and the generic name refer to the same molecule.
MC4R is generally described as the principal target, based on binding affinity and functional assays. Activity at MC1R, MC3R and MC5R is reported as weaker. Receptor selectivity is one reason the compound was pursued for central rather than vascular effects.
Both are synthetic melanocortin analogues built on a similar cyclic peptide scaffold. Bremelanotide is described in the literature as a metabolite-derived analogue of melanotan II rather than the same substance. The two differ in terminal modifications, which affect half-life and receptor profile.
The peptide is polar and charged, and reversed-phase chromatography with acidic mobile phases resolves it well from related substances. Ultraviolet detection near 214 nm gives adequate sensitivity without derivatisation.
